An Alternate Method for Staining Plastic Embedded Autoradiographs

Abstract

We have developed a rapid and reliable method for staining plastic embedded autoradiographs for in situ labeling protocols that eliminates several problems sometimes encountered with other methods, such as displacement of silver grains and uptake of stain by the emulsion layer itself. The procedure features the brief (less than one minute) application of toluidine blue to plastic embedded autoradiographs with moderate warming. The quality of staining obtained by this method is both consistent and flexible, allowing varying degrees of intensification without the danger of damaging the emulsion layer. The principle advantages of this procedure over other techniques to stain plastic embedded autoradiographs are its simplicity, rapidity and reproducibility. (The J Histotechnol 14:43, 1991)
