Near-field microscopy of living cells

Abstract

Near-field optical microscopy (NSOM) coupled with other techniques such as fluorescence, is a promising tool for high-resolution imaging of the molecular organization of cell membranes. A NSOM designed to maintain living cells in buffer is presented. Both fixed and live cells in liquid have been imaged, using fluorescent antibodies, lectins, and lipid analogues as probes of membrane organization.
