Abstract
The development of new technologies capable of monitoring the delicate details of the dynamics of living cells, which remain outside the optical resolution, is vital for cell research. The infrared light introduces several unique advantages for cell studies. Its interaction with live tissue causes no photodamage and allows examining chemical information encoded in infrared absorption spectrum. This chapter presents newly developed infrared surface plasmon resonance (SPR) methodology. This chapter discusses a specific aspects related to studies of aqueous samples using infrared surface plasmon. The extreme sensitivity of the surface plasmon to refractive index changes together with extended probing depth in the infrared range in comparison to visible range allows studying living cell morphology with submicron precision. Analysis of the temporal variation of the SPR reflectivity allows reconstructing the dynamics of the cell shape and observing various stages in cell behavior. Using infrared SPR technique we have studied the dynamics of cellular processes and successfully monitored submicron changes in cell morphology.
