Use of the saturation curves for evaluating distances in biological specimens by the method of double spin labels

Abstract

A method is proposed for determining the distances from the functional groups of protein to the paramagnetic metal in the active centre based on the plotting of the saturation curves of the EPR spectra of spin-labelled proteins. The saturation curves proved to be more sensitive to the interaction with the rapidly relaxing paramagnetic centre (p.c.) than the width and form of the line. Analysis of the saturation curves in certain conditions makes it possible to identify various types of labels differing in the value of interaction with the p.c. and also to evaluate the depth of the submergence of the p.c. in the protein globule.
